Product Description
Human Umbilical Vein Endothelial Cells (HUVECs) were isolated from normal human umbilical cord tissue. These cells were genetically modified via lentiviral transduction to stably express VE-Cadherin-GFP, followed by puromycin selection. This allows live visualization of cell junction dynamics and vascular integrity in real time. Cells are provided at passage 1 and shipped frozen. We recommend using Nuvyra Labs Endothelial Growth Medium for optimal cell performance.

Handling Upon Arrival
Transfer vials immediately to a −80°C freezer for short-term storage or to a liquid nitrogen tank for long-term preservation upon delivery.
Thawing & Subculturing Instructions
A) Pre-coat a T25 flask with 2 mL Coating Solution, incubate for 5 min, rinse with 4 mL rinse solution, and aspirate to prepare.
B) Thaw the vial in a 37°C water bath, disinfect the exterior with 70% ethanol, and open in a biosafety cabinet. Seed into flask with 10 mL Endothelial Growth Medium.
C) Once confluent, rinse twice with 5 mL rinse solution, add 2 mL Detachment Solution, aspirate excess after 20 seconds.
D) Incubate at RT or 37°C for 1–2 min until cells round up; tap flask gently to dislodge cells.
E) Add 5 mL Neutralization Solution, centrifuge at 800g for 5 min.
G) Resuspend pellet with 10–15 mL medium and split into 2–3 pre-coated flasks.
H) Refresh medium every 2–3 days; confluence typically reached in 7 days at 1:3 ratio.
I) For quiescent conditions, switch to Basal Medium for 12 hrs prior to experiments.
