Product Description
Human Umbilical Vein Endothelial Cells (HUVECs) were isolated from healthy human umbilical cord tissue. These primary endothelial cells are commonly used in vascular biology studies, including angiogenesis, permeability, and inflammatory responses. Cells are cryopreserved at passage 1 and shipped in frozen vials. Use of Nuvyra Labs Endothelial Growth Medium is recommended for optimal performance.

Handling Upon Arrival
Upon receiving the dry ice shipment, immediately store frozen vials in a −80°C freezer (short term) or transfer to liquid nitrogen (long term).
Thawing & Subculturing Instructions
A) Pre-coat T25 flasks with 2 mL Coating Solution, incubate for 5 min, rinse with 4 mL Rinse Solution, and remove.
B) Thaw cells in a 37°C water bath, sanitize vial with 70% ethanol, and work in a biosafety cabinet. Add thawed contents to a pre-coated T25 flask with 10 mL Endothelial Growth Medium.
C) When confluent, rinse twice with 5 mL Rinse Solution, add 2 mL Detachment Solution, and aspirate excess within 20 seconds.
D) Incubate at RT or 37°C for ~1–2 min until cells round up. Tap gently to detach cells.
E) Add 5 mL Neutralization Solution and centrifuge at 800g for 5 min.
G) Resuspend in 10–15 mL medium and seed 5 mL/flask into 2–3 flasks (1:2 or 1:3 ratio).
H) Replace medium every 2–3 days. Expect confluence in ~7 days.
I) To prepare quiescent cells, switch to Basal Medium for ~12 hours before experiments.
