Handling Upon Arrival
Transfer frozen vials immediately to −80°C (short term) or liquid nitrogen (long term). Handle with appropriate PPE.
Thawing & Subculturing Instructions
A) Pre-coat T25 flask with 2 mL Coating Solution, incubate 5 min, rinse with 4 mL rinse solution, and aspirate.
B) Thaw vial in a 37°C water bath, disinfect, and transfer to biosafety cabinet.
C) Add 10 mL Endothelial Growth Medium to flask and seed cells.
D) Once confluent, rinse flask twice with 5 mL rinse solution, add 2 mL Detachment Solution, aspirate after 20 seconds.
E) Wait 1–2 minutes at RT or 37°C, then tap flask to dislodge cells.
F) Add 5 mL Neutralization Solution, centrifuge at 800g for 5 min.
G) Resuspend pellet in 10–15 mL medium, seed into 2–3 flasks (1:2 to 1:3 split).
H) Change medium every 2–3 days; cells typically reach confluence within 7 days.
I) For quiescent cells, switch to Endothelial Basal Medium for 12 hrs before experiments.
Cells are offered for Research Use Only. Not for Clinical Use.
