Handling Upon Arrival
Upon delivery on dry ice, immediately store in −80°C for short term or in liquid nitrogen for long-term use. Do not thaw unless preparing for culture.
Thawing & Subculturing Instructions
A) Pre-coat a T25 flask with 2 mL Nuvyra Labs Coating Solution, incubate 5 min, rinse with 4 mL cell rinse solution, remove and discard rinse.
B) Thaw vial in 37°C water bath, disinfect, and transfer to biosafety cabinet.
C) Add 10 mL Endothelial Growth Medium to the flask and seed cells.
D) When confluent, subculture:
-
Rinse cells with 5 mL rinse solution ×2
-
Add 2 mL Detachment Solution, aspirate excess
-
Wait 1–2 min at RT or 37°C, tap flask to dislodge cells
-
Add 5 mL Neutralization Solution, centrifuge at 800g for 5 min
E) Resuspend pellet in 10–15 mL medium, split into 2–3 pre-coated flasks
F) Change medium every 2–3 days; cells reach confluence in ~7 days
G) To induce quiescence, replace with Endothelial Basal Medium for 12 hrs before experiment
Cells are offered for Research Use Only. Not for Clinical Use.
