Handling Upon Arrival
Thawing & Subculturing Instructions
A) Pre-coat T25 flask with 2 mL Nuvyra Labs Coating Solution, incubate 5 min, rinse with 4 mL cell rinse solution.
B) Thaw vial in 37°C water bath, disinfect exterior with 70% ethanol, transfer contents into biosafety cabinet.
C) Seed into flask with 10 mL Endothelial Growth Medium.
D) For passaging:

Rinse with 5 mL rinse solution (×2)

Add 2 mL Detachment Solution, aspirate excess in 20 seconds

Wait 1–2 min at RT or 37°C
-
Tap flask to detach
-
Neutralize with 5 mL Neutralization Solution

Centrifuge at 800g × 5 min
E) Resuspend and seed into 2–3 pre-coated flasks (1:2 or 1:3 ratio).
F) Medium change every 2–3 days.
G) For quiescent cells, switch to Endothelial Basal Medium for 12 hrs before experiments.
