Thawing & Subculturing Instructions
-
Pre-coat flask: Add 2 mL Nuvyra Labs Coating Solution to a T25 flask. Incubate 5 min. Aspirate and rinse with cell rinse solution.
-
Thawing: Warm vial in 37°C water bath. Disinfect with 70% ethanol. Transfer cells into biosafety cabinet.
-
Seeding: Add 10 mL Nuvyra Labs Endothelial Growth Medium to pre-coated flask and seed cells.
-
Passaging:
-
Rinse flask twice with rinse solution
-
Add 2 mL Detachment Solution, aspirate after 20 sec
-
Wait 1–2 min, gently tap to dislodge cells
-
Add 5 mL Neutralization Solution, centrifuge 800g, 5 min
-
Resuspend and seed into 2–3 pre-coated flasks (1/2 to 1/3 ratio)
-
Feeding: Change medium every 2–3 days
-
Quiescence: When confluent, switch to Endothelial Basal Medium 12 hours before experiments
Note: Performance is only guaranteed with Nuvyra Labs reagents.
