Handling Instructions Upon Arrival
Transfer the frozen vial to a −80°C freezer for short-term storage or to a liquid nitrogen tank for long-term preservation.
Thawing and Subculturing Protocol
A) Pre-coat T25 flasks with 2 mL Nuvyra Labs Coating Solution, incubate 5 minutes. Aspirate and rinse with 4 mL rinse solution.
B) Thaw vial in 37°C water bath, sanitize, and transfer cells into a biosafety cabinet. Add 10 mL Nuvyra Labs Endothelial Growth Medium to the pre-coated flask.
C) Once confluent, rinse twice with 5 mL rinse solution. Add 2 mL Nuvyra Labs Detachment Solution, aspirate after 20 seconds.
D) Incubate at RT or 37°C for 1–2 minutes. Tap flask to detach cells.
E) Add 5 mL Nuvyra Labs Neutralization Solution and centrifuge at 800g for 5 minutes.
F) Resuspend cells in 10–15 mL medium, plate 5 mL into 2–3 pre-coated T25 flasks.
G) Replace medium every 2–3 days. Cells typically reach confluence within 7 days (1/3 split ratio).
H) For quiescent cultures, switch to Nuvyra Labs Endothelial Basal Medium for ~12 hours prior to experiments.
