Thawing & Subculturing Protocol

  1. Pre-Coat Flask:
Add 2 mL of Nuvyra Labs Coating Solution to a T25 flask. Incubate for 5 min. Aspirate and rinse with 4 mL Cell Rinse Solution. Flask is now ready.

  2. Thawing:
Thaw vial in a 37°C water bath. Wipe with 70% ethanol and transfer to biosafety cabinet. Add 10 mL of Endothelial Growth Medium to pre-coated flask and seed cells.

  3. Passaging:

    • Rinse flask twice with 5 mL Cell Rinse Solution.

    • Add 2 mL Detachment Solution. Aspirate after 20 seconds.

    • Incubate 1–2 min at RT or 37°C. Tap flask if needed for detachment.

    • Add 5 mL Neutralization Solution, centrifuge at 800g for 5 min.

  4. Seeding:
Resuspend in 10–15 mL medium. Plate 5 mL per T25 flask (1:2 or 1:3 ratio).
Change medium every 2–3 days. Confluence expected in ~7 days.

  5. For Quiescent Cells:
When nearly confluent, switch to Endothelial Basal Medium for 12 hours before experiments.
