Product Description
Human Brain Microvascular Endothelial Cells (HBMVECs) were isolated from normal human brain cortex tissue. Cells were genetically modified by lentiviral transduction to express VE-CAD-GFP and subsequently selected with puromycin. Cells are provided at passage 1 and shipped frozen. Nuvyra Labs Endothelial Growth Medium is strongly recommended for culturing these cells.

Thawing and Subculturing Instructions

  1. Pre-Coating: Add 2 mL of Nuvyra Labs Coating Solution to a T25 flask, incubate 5 min at RT. Rinse with 4 mL cell rinse solution.

  2. Thawing: Thaw vial in a 37°C water bath. Wipe with 70% ethanol. Transfer to biosafety cabinet.

  3. Seeding: Add 10 mL Nuvyra Labs Endothelial Growth Medium to flask and add thawed cells.

  4. Passaging: Rinse flask with 5 mL rinse solution twice. Add 2 mL detachment solution. After 1–2 minutes, tap flask gently to release cells.

  5. Neutralization: Add 5 mL Nuvyra Labs Neutralization Solution. Centrifuge at 800 × g for 5 minutes.

  6. Expansion: Resuspend in 10–15 mL medium. Seed 5 mL per pre-coated T25 flask (1:2–1:3 ratio).

  7. Medium Change: Every 2–3 days. Confluence typically reached in 7 days.

  8. Quiescence Preparation: Switch to Nuvyra Labs Endothelial Basal Medium for 12 hours prior to experiments when cells are nearly confluent.
