Thawing and Subculturing Protocol

  1. Pre-Coating Flask
Add 2 mL Universal Coating Solution to a T25 flask. Incubate at room temperature for 5 minutes. Discard excess solution and rinse with 4 mL of Cell Rinse Solution. Remove PBS and proceed to seeding.

  2. Thawing Cells
Thaw the frozen vial in a 37°C water bath. Wipe the vial with 70% ethanol before transferring to a biosafety cabinet. Seed cells into a pre-coated T25 flask containing 10 mL of Nuvyra Labs Endothelial Growth Medium.

  3. Passaging

    • Rinse with 5 mL Cell Rinse Solution twice.

    • Add 2 mL Nuvyra Labs Detachment Solution and aspirate excess after 20 seconds.

    • Incubate at RT or 37°C for 1–2 minutes. Tap flask gently to dislodge cells.

    • Add 5 mL Nuvyra Labs Neutralization Solution. Centrifuge at 800g for 5 minutes.

  4. Expansion

    • Resuspend pellet in 10–15 mL medium.

    • Transfer 5 mL into each of 2–3 pre-coated T25 flasks (1:2–1:3 ratio).

    • Change medium every 2–3 days. Cells typically reach confluency in 7 days.

  5. Preparing Quiescent Cells
When nearly confluent, switch to Nuvyra Labs Endothelial Basal Medium for 12 hours before experiments.
