Product Description
Human Umbilical Vein Endothelial Cells (HUVECs) were isolated from healthy human umbilical cord tissue and genetically modified using lentiviral vectors to express Chimeric GFP-Aequorin localized to mitochondria. These HUVEC-AEQ-Mito cells are puromycin-selected and ideal for live-cell mitochondrial calcium imaging, bioenergetics studies, or calcium signaling research. Cells are provided at passage 1 and shipped cryopreserved. Use Nuvyra Labs Endothelial Growth Medium for optimal results.

Handling Upon Arrival
Transfer vials to a −80°C freezer immediately upon arrival for short-term storage or to a liquid nitrogen tank for long-term storage.
Thawing & Subculturing Instructions
A) Pre-coat T25 flasks using 2 mL Coating Solution, incubate 5 min, rinse with 4 mL rinse solution, aspirate, and prepare for seeding.
B) Thaw cells in a 37°C water bath, disinfect with 70% ethanol, and transfer under a biosafety cabinet. Seed into 10 mL Endothelial Growth Medium in prepared flask.
C) Once confluent, rinse twice with 5 mL rinse solution, add 2 mL Detachment Solution, and aspirate excess after 20 seconds.
D) Incubate 1–2 min until cells round up. Tap gently to dislodge.
E) Add 5 mL Neutralization Solution, centrifuge at 800g for 5 min.
G) Resuspend in 10–15 mL medium, distribute 5 mL each into 2–3 T25 flasks (split 1:2 or 1:3).
H) Refresh medium every 2–3 days. Cells typically reach confluence in 7 days.
I) To prepare quiescent cells, switch to Basal Medium for 12 hours prior to experiments.
