Transfer frozen vials to a −80°C freezer (short-term) or liquid nitrogen tank (long-term).

Thawing and Subculturing Protocol

A) Pre-coat T25 flasks with 2 mL of Nuvyra Labs Coating Solution, incubate 5 minutes at room temperature. Aspirate and rinse with 4 mL of cell rinse solution. Discard PBS; flask is ready.

B) Thaw the vial in a 37°C water bath. Spray with 70% ethanol and wipe before opening in a biosafety cabinet. Add 10 mL of Nuvyra Labs Endothelial Growth Medium to a pre-coated flask and transfer the cell suspension.

C) To passage cells:

  • Rinse with 5 mL of cell rinse solution twice.

  • Add 2 mL of Nuvyra Labs Detachment Solution and aspirate within 20 seconds.

  • Leave at RT or 37°C for 1–2 minutes. Tap flask to release cells.

D) Add 5 mL of Nuvyra Labs Neutralization Solution, centrifuge at 800 × g for 5 minutes.

E) Resuspend the pellet in 10–15 mL medium and seed 5 mL into 2–3 pre-coated T25 flasks (1/2 to 1/3 subculture ratio).

F) Change medium every 2–3 days. Cells typically reach confluence within 7 days.

G) For quiescent cells, replace growth medium with Nuvyra Labs Endothelial Basal Medium for ~12 hours before experiments.

Important Notes

  • Human tissue-derived products are potentially biohazardous. Follow standard biosafety precautions.

  • Use only Nuvyra Labs reagents to ensure growth and marker fidelity.

  • Quality concerns must be reported within one month of delivery. Replacements are not guaranteed beyond that period.
